Culture of Peripheral Blood Leucocytes for Chromosome Analysis Using a Few Drops of Whole Blood.

چکیده
A number of modifications of Moorhead's original technique for culturing peripheral leucocytes have been described, enabling cultures to be grown from a few drops of blood (Arakaki and Sparkes, I963; Edwards, I962; Robinson, Bishun, Rashad, and Morton, I964). They require the use of stored calf, chick, or human serum, and this has obvious disadvantages for the small laboratory. We have, therefore, recently been using a technique for culturing small amounts of blood from a baby using plasma from a parent or other convenient adult. Three or four drops of blood can be collected from the baby from a heel prick. They can be conveniently collected into a disposable syringe containing o5 ml. Glaxo i99 medium and ioo units heparin. If the mixture is transferred to a bottle it can be kept for up to an hour at least. A io-ml. blood sample is taken from the parent, and some of this is used to set up a culture in the usual way. The residue is then spun quickly, until i.5 ml. clear plasma can be removed. To ensure

Induction of premature chromosome condensation (PCC) by calyculin A for biodosimetry
Background: Premature chromosome condensation (PCC) is a method for interphase chromosome analysis in biodosimetry. This study was performed to verify the usefulness of PCC induced by calyculin A in human peripheral blood lymphocytes (pbl) for biological dosimetry and possible construction of dose-response curve. Materials and Methods: Peripheral blood was obtained from a healthy donor and expo...
متن کاملBiological Dose Assessment by Chemical Induction of Premature Chromosome Condensation
Dose assessment of radiation victims is a key element in medical management of radiation accidents. At high radiation doses (> 6 Gy), dicentric assay lose its efficiency and premature chromosome condensation (PCC) assay was proposed to overcome the restrictions of dicentric assay. It was shown that PCC ring is more suitable and simpler than dicentric for biodosimetry in high radiation doses. In...
متن کاملActivity of T-Helper Lymphocytes in Opium Dependent and Non-dependent Individuals
Opium derivatives affect the function and differentiation of peripheral leucocytes, directly or indirectly. Previous researches have shown that these cells respond to the endorphins through their opioid receptors (σ, µ and κ). There are many reports about the effect of morphine and heroin on immune system of dependent people, but there is no report on the functions of T-helper lymphocytes in op...
متن کاملA multi-step centrifuge method for extraction of leucocytes and preparation of Tc-HMPAO labeled leucocytes
Background: Labeled leucocytes could be used for localization of infection foci after surgeries or in inflammatory diseases including inflammatory bowel diseases. Extraction of leucocytes needs 10% Hetastarch which is not available in Iran. We provide a method employing multiple centrifuges to extract and label leucocytes with Tc-HMPAO. Methods: The study was conducted from April to June 2018 ...
متن کاملInterindividual differences in radiation-induced apoptosis of peripheral blood leukocytes in normal individuals and breast cancer patients
Background: Quantification of radiation-induced apoptosis in peripheral blood lymphocytes (PBLs) has been proposed as a possible screening test for cancer-prone individuals and also for the prediction of normal tissue responses after radiotherapy. Materials and Methods: The neutral version of the comet assay (single-cell gel electrophoresis) was used 24, 48, 72 hours after irradiation ...
